Freeze-drying/lyophilization: What you need to know

Procedure

Frostung/Gefrierung:
Die Produkte werden in einer Vorrichtung auf den Temperaturplatten platziert und werden durch die Oberflächentemperatur der Platte auf unter -18 °C herunter gekühlt. Sobald die Gefriertemperatur erreicht ist, wird die Anlagenkammer unter Vakuum gesetzt. Meist wird hier ein Vakuum von unter 200 mbar benötigt.
Primärtrocknung/Sublimation:
In diesem Prozessabschnitt wird die Temperatur der Platten erhöht. Abhängig vom Produkt kann die Temperatur hier auf 0 °C bis 15 °C ansteigen. Durch die sehr langsame Erwärmung der Platten wird dem Produkt die Flüssigkeit entzogen, ohne dabei zu verflüssigen. Die Primärtrocknung ist der wichtigste und zugleich kritischste Prozessabschnitt in einer Gefriertrocknung.
Sekundär-/Nachtrocknung:
In der Sekundärtrocknung wird abschließend das stark gebundene Wasser durch eine weitere Erhöhung der Temperatur auf über +50 °C in Wasserdampf umgewandelt, um den Wassergehalt auf 0,5 – 3 % zu senken.
